Summary
Freund's Complete Adjuvant (FCA/CFA) is carefully formulated with reference to the classic preparation method from paraffin oil and other low-endotoxin, high-purity oils that cannot be metabolized by animals, and inactivated BCG. It does not contain preservatives. It is one of the most commonly used immune adjuvants and is often used to prepare antibodies for animal immunization.
Freund's complete adjuvant is Freund's incomplete adjuvant with the addition of Mycobacterium tuberculosis. Freund's complete adjuvant has a stronger ability to enhance immune responses and is generally used for primary immunization. Freund's incomplete adjuvant is generally used for booster immunization after the initial immunization, which can avoid the side effects of Freund's complete adjuvant and facilitate the production of high-titer antibodies.
The mechanism of action of adjuvants is not fully understood. Adjuvants may increase the surface area for antigen presentation, making it easier for macrophages to phagocytose; they may also prolong the persistence of antigens in the body and increase the chance of contact with immune cells; they may also enhance the ability of antigen-presenting cells to present and process antigens; Or it can induce an inflammatory reaction at the antigen injection site and its local lymph nodes, thereby stimulating the proliferation of immune cells. All of the above mechanisms may increase the titer of antibodies produced by the body during primary immunization and booster immunization.
