Cholinesterase Classification
Cholinesterase can generally be divided into two categories:
 
true cholinesterase
Distributed in red blood cells and brain gray matter. True cholinesterase, also known as acetylcholinesterase (AChE), mainly acts on acetylcholine and has high specificity
 
pseudocholinesterase
Distributed in liver, brain white matter and serum. Pseudocholinesterase, also known as pseudocholinesterase, includes propionylcholinesterase (PrChE), benzoylcholinesterase (Benzoylcholinesterase, BzChE), butyrylcholinesterase (BuChE) ), usually the main one in serum is butyrylcholinesterase.
 
 
Cholinesterase detection methods and principles
 
DGKC Method
Cholinesterase catalyzes the hydrolysis of butyrylthiocholine to produce butyrate and thiocholine. Thiocholine reduces yellow potassium hexacyanoferrate (Ⅲ) (potassium ferricyanide) into colorless potassium hexacyanoferrate (Ⅱ) (potassium ferrocyanide), and the absorbance decreases at 405nm. Monitored continuously, the rate of decline is directly proportional to the cholinesterase activity in the sample.
 
DTNB Method
The enzyme reaction product, thiocholine, reacts with 5,5′-dithiobis(2-nitrobenzoic acid) (DTNB) to generate 5-mercapto-2-nitrobenzoic acid. This appears yellow at a certain pH, and the color intensity is measured colorimetrically. This method is highly sensitive, requires a small amount of tissue, does not terminate the enzyme reaction, and is suitable for continuous observation of enzyme activity.
 
Dry Chemistry
The enzyme reaction product, thiocholine ester, reacts with DTNB to generate 5-mercapto-2-nitrobenzoic acid. This appears yellow at a certain pH, and the color intensity is measured colorimetrically. This method is highly sensitive, requires a small amount of tissue, does not terminate the enzyme reaction, and is suitable for continuous observation of enzyme activity.
