Molecular diagnostic technology refers to the use of DNA and RNA as diagnostic materials. Through molecular biology techniques, it detects the presence, defects, or abnormal expression of genes. This allows for the diagnosis of the individual's health status and diseases. The basic principle involves examining whether there are structural changes in DNA or RNA, the quantity of these molecules, and whether their expression functions are abnormal. This helps determine whether the individual has any abnormal changes at the genetic level, which is of crucial importance for the prevention, prediction, diagnosis, and prognosis of diseases. In simple terms, all methodological techniques based on the molecular biology level are considered part of molecular diagnostic technology, such as PCR technology, gene sequencing, and more.
Polymerase Chain Reaction (PCR) is a molecular biology technique used to amplify specific nucleotide segments. It can be viewed as a special nucleic acid replication that occurs outside a living organism. PCR is characterized by its ability to significantly amplify minute amounts of nucleic acids, making them easily identifiable and distinguishable.
PCR Technology
PCR can amplify target DNA segments more than a million times. The principle involves using DNA polymerase to catalyze the process, using the parent DNA strand as a template and specific primers as extension starting points. Through denaturation, annealing, extension, and other steps, PCR replicates the process of generating a complementary DNA strand from the parent DNA template outside the living organism.
The standard PCR process consists of three steps:
Denaturation: Utilizing high temperature to separate the DNA double strands. The hydrogen bonds between the DNA double strands are disrupted at elevated temperatures (93-98°C).
Annealing: After the DNA double strands are separated, the temperature is lowered, allowing primers to bind to the single-stranded DNA.
Extension: DNA polymerase starts synthesizing the complementary strand along the DNA chain from the primer-binding site during the temperature drop. Once extension is complete, one cycle is finished, and the number of DNA fragments doubles.
Repeating these three steps 25-35 times exponentially increases the number of DNA fragments.
