Guanidine Isothiocyanate in RNA Extraction
 
TRIzol is a comprehensive RNA extraction reagent that can directly isolate total RNA from cells or tissues. Key components of TRIzol include guanidine isothiocyanate and phenol. Guanidinium isothiocyanate effectively lyses cells, facilitates ribosomal dissociation, separates RNA from proteins, and releases RNA into the solution.
 
When chloroform is introduced, it extracts the acidic phenol, leading to the partitioning of RNA into the aqueous phase. Following centrifugation, the aqueous layer (colorless) primarily contains RNA, while the organic layer (yellow) consists primarily of DNA and protein.
 
 
 
 
Applications of Guanidine Isothiocyanate
 
RNA extraction from tissue cells and serum samples, ideal for denaturing nucleic acids and isolating RNA;
As a storage buffer for whole blood samples;
In RNA lysis stabilization buffer formulations, used to lyse colorectal adenocarcinoma cell lines prior to RNA isolation;
Inactivates nucleases, excellent for storing and freezing stool samples for DNA research;
As a PCR inhibitor in mitochondrial DNA (mtDNA) qPCR reaction systems;
For qPCR analysis of DNA extracted from cerebrospinal fluid (CSF).
