Diethylpyrocarbonate (DEPC) is a chemical employed to deactivate RNase enzymes and is susceptible to moisture and pH fluctuations. In aqueous solutions at 155°C, it can break down into ethanol and carbon dioxide. DEPC is also responsive to ammonia, which can transform it into urethane, a potential carcinogen.
 
DEPC acts as a potential nuclease inhibitor, with a 0.1% DEPC solution commonly used for RNase inactivation. It hinders the binding of ryanodine to ryanodine/Ca2+ receptor channels in skeletal muscle in a dose- and time-dependent manner, increasing the Ca2+ permeability of SR vesicles. At a concentration of approximately 0.1% (v/v) in solution, it effectively deactivates RNase, thereby preventing RNA degradation.
 
Diethyl pyrocarbonate finds application in treating deionized water in PCR reactions to minimize the risk of RNA degradation by RNase. Additionally, it is suitable for dot blot hybridization to dilute total RNA extracted from various microorganisms.
 
As a modification reagent, DEPC targets His and Tyr residues in proteins. It serves as a robust probe for disrupting dsDNA structure by reacting with unstacked bases.
