Leucine aminopeptidase (LAP) is a protease found extensively in various human tissues. Employing L-Leucine-p-nitroanilide as the substrate, LAP undergoes hydrolysis, and the LAP activity is assessed by detecting P-nitroaniline. This process proves valuable in diagnosing liver tract obstruction and pancreatic cancer.


Overview of LAP

Leucine aminopeptidase (LAP) is a proteolytic enzyme that catalyzes the hydrolysis of the N-terminus of peptide chains, forming peptide bonds between leucine and other amino acids. LAP is widely distributed in tissues such as the liver, pancreas, and kidneys. It serves as a valuable diagnostic tool for conditions like liver tract obstruction and pancreatic cancer. Unlike other liver function enzymes limited to blood sample testing, LAP can also be assessed in urine.
The substrate employed is L-leucine p-nitroaniline hydrochloride, and LAP activity is determined by detecting p-nitroaniline (P-nitroaniline) following its hydrolysis by LAP.


LAP Detection Substrate

Measurement principle:LAP hydrolyzes the substrate to produce p-nitroaniline, and measures the absorbance per minute of p-nitroaniline at a wavelength of 405nm
Change rate (ΔA/min), calculate the activity of LAP in the sample to be tested.
Advantages:The test is fast, easy to operate, and suitable for fully automatic biochemical analyzers.
